In vitro Multiple Shoot Regeneration in Carnation (Dianthus caryophyllus L.)

Abastract
Multiple shoot regeneration was tried from nodal and shoot tip explants of carnation (Dianthus caryophyllus L.) on MS and modified half MS medium containing different concentrations of BAP, NAA and Kn. Of the different combinations of BAP and Kn used, best response was achieved in half MS medium containing 1.5 mg/1 BAP and 0.5 mg/1 Kn. In this combination 10-12 shoot buds were initiated from a single explant and the number of multiple shoots increased at least two- threefold within 10-14 days when they were subcultured in the same medium. On the other hand, shoot tip explant produced 8-9 shoots per explant in MS medium containing 0.75 mg/1 BAP and 0.2 mg/1 NAA. These regenerated multiple shoots were devoid of roots. Best response towards healthy rooting was achieved on 0.2 mg/1 IBA and 0.2 mg/1 NAA used in MS medium solidified with phytagel.
