In vitro multiple shoot regeneration in Chrysanthemum morifolium Ramat

Abastract
Different concentrations of IAA, NAA, BAP and Kn were used singly or in various combinations on MS medium to optimize multiple shoot regeneration from shoot tip and nodal explants of Chrysanthemum morifolium Ramat. Multiple shoot regeneration was observed from both explants when they were cultured on MS medium supplemented with either BAP or in combinations with IAA, NAA and Kn. Among the two explants, shoot tips showed comparatively better response to multiple shoot regeneration. Although cent per cent response to multiple shoot regeneration was achieved on MS medium supplemented with IAA and BAP , the maximum number of multiple shoot regenerated from shoot tip explants when 1.0 mg/1 BAP + 1.0 mg/1 NAA were added to MS medium. In this composition, 20-22 shoot buds were initiated from a single explant. Of the various concentrations of IBA used for root induction, the maximum response was obtained on MS medium containing 0.2 mg/1 IBA. The regenerated shoots with well-developed roots were successfully transferred into small pots; and after proper hardening they were established into clay pots.
