MORPOGENIC RESPONSES OF PEANUT LEAFLET EXPLANTS CULTURED IN VITRO.

Leaflet explants of peanut were cultured on modified MS medium containing different concentrations and combinations of 2,4-D, NAA, Kn and BAP for callusing and shoot production. NAA and 2,4-D gave good callus but no shoots. Shoot buds were obtained when BAP was used either singly (0.5, 1.0, 1.5, 2.0 and 2.5 mg/1) or in combination with kinetin and NAA (BAP 2mg/1, Kn 0.5 mg/1 and NAA 1.0 mg/1). Histological studies of differentiating callus showed the presence of meristemoids that developed into green buds and shoots. Regenerated shoots produced profuse roots when they were cultured in half strength MS medium containing 0.2 mg/1 IBA.
