EFFECT OF pH LEVEL AND SUCROSE CONCENTRATION ON GRAFT FORMATION IN SOYBEAN INTERNODES IN VITRO

The effect of pH level and sucrose concentration in the culture medium containing 2.15 mg/1 kinetin and 20.00 mg/1 IAA on graft development has been studied using excised internodes of Glycine max in vitro. A pH range of 5.5 to 6.0 and sucrose concentration of 2.0% in the culture medium were found to be the most effective in xylem differentiation in leading to the development of graft union. A higher pH of 6.5 and sucrose concentration of either 1.0 or 3.0% in the culture medium were found to affect callus growth and xylem differentiation as well as graft development in vitro.
