IN VITRO CULTURE OF CARICA PAPAYA L. III. MAINTENANCE PROLIFERATING CULTURE

In vitro culture of Carica papaya L. cv. ?Rajshahi-red? was initiated by culturing shoot apices from mature plant onto MS medium supplemented with 10 mg/1 2ip + 1.0 mg/1 NAA and multiple lateral bud proliferation was induced after transferring into half the strength of MS medium (4 weeks) having 0.1 mg/1 BA + 0.02 mg/1 NAA. Effects of different subculture on proliferating culture was studied by : (i) individual buds from proliferating culture directly transferred to next subculture (sbm-1) and (ii) individual buds before transferring to the proliferation medium of each subculture were incubated 4 weeks on hormone free half the strength of MS medium (sbm-2). Individual bud culture maintained through sbm-1 showed maximum regenerability up to 7th subculture but proliferation ability and rooting capacity gradually decreased from 8th subculture and finally lost at 11th subculture. However, subculture of individual buds through sbm-2 showed more or less constant regenerability up to 30th subculture.
