A non-commercial, direct current square wave generator was used to induce the uptake of plasmid into plant protoplasts. High levels of transient gene expression were obtained without severe loss of protoplast viability in the leguminous species Glycine argyrea, Medicago sativa, M. varia and Stylosanthes macrocephala.
