In vitro Plant Regeneration from Leaf-derived Callus Cultures of Adhatoda vasica Nees

Abastract
The young leaf explants from the in vitro established shoot cultures of mature plant origin were induced to from callus subsequently to differentiate into shoots on MS medium. The cultured explants produced calli from their cut margins within two weeks and shoot buds within three weeks of incubation on media supplemented with 0.5-2.0 mg/1 BA and 0.1-0.5 mg/1 NAA. Maximum number of shoots with optimum callus growth was observed in cultures grown on MS medium containing 1.0 mg/1 BA and 0.1 mg/1 NAA. Rooting was induced in in vitro regenerated shoots on half strength on MS medium with 0.1-05 mg/1 of either NAA or IBA, or without any growth regulator. The best rooting was observed when the microcuttings were rooted on hormone-free medium. The in vitro regenerated plantlets were successfully established on the soil.
