Efficient in vitro Shoot Propagation of Wild Species of Oryza and Effect of Different Gelling Agents

Abastract
Single shoots from proliferated shoots of various wild species of Oryza were cultured on MS medium containing 3 per cent sucrose, supplemented with different levels of BAP or Kn and solidified with either agar or Phytagel for shoot proliferation. After 30 and 60 days of culture, proliferated shoots were counted. In general, media supplemented with BAP were found to be superior to those supplemented with Kn for rapid propagation of all wild species examined. The optimum level of BAP for rapid multiplication of shoots ranged from 4.0-10.0 mg/1 and 6.0-10.0 mg/1 for Kn. The effect of solidifying agents on plant proliferation was also observed. Shoots cultured on Phytagel media showed high rates of multiplication than agar solidified media in all the species. Such a simple technique of efficient clonal propagation could provide a sufficient number of hybrid material for wide hybridization and callus induction for certain applications of biotechnological technique.
