Micropropagation of Gladiolus Through In Vitro Cormlets Production

Abastract
The flower stalk explants were investigated for establishing in vitro propagation of Gladiolus in three cultivars viz. Mayur, Sylvia, Spic and Span. The nodal segments responded better than internodal-, rachis-and floret-segments irrespective of the genotype and culture medium. Nodal segments regenerated single shoot per explant upon culturing on MS medium supplemented with BAP (1 mg/1), whereas, those cultured on MS + BAP (5 mg/1) medium developed 14-20 shoot primordia within 4 weeks. The shoot primordia clump sections (4-6 buds) were multiplied 5-6 fold through subculturing (4-5 weeks) on the same fresh medium. The best shoot regeneration from these multiple shoot-bud-clumps was obtained by culturing on MS basal medium. The shoots individually cultured in liquid half strength MS + IBA (4 mg/1) + sucrose (6 or 9%) developed cormlets (3-13 mm diameter) within 4-5 weeks. The mature cormlets developed in vitro collected from the test tubes.
