Micropropagation of Cymbidium bicolor through in vitro Culture

Abastract
Five different media, namely, Phytamax, Modified Vacin and Went (MVW), Knudson C (KC), modified Knudson C (MKC) and Lindemann with various supplements and additives were tested for large scale multiplication of Cymbidium bicolor via seeds . Of the five media tried Phytamax was found to be the best for seed germination and protocorm development. The second highest response was recorded in MVW and in Lindemann the response was least. Seed germination was optimum when Phytamax medium was supplemented with 2.5 mg/1 BAP + 1.0 mg/1 Kn. The seeds germinated within 35-40 days. Protocorn development was studied in both agar solidified and liquid Phytamax media. Although a large number of protocorms were multiplied in both solid and liquid media, the growth and development of the protocorns were more vigorous and faster in liquid medium. Roots developed within 12-22 days and the medium devoid of any hormonal supplements was found best.
