HISTOLOGY OF IN VITRO ORGANOGENESIS IN GUAVA CULTURES ESTABLISHED FROM SHOOT EXPLANTS OF MATURE TREES

Histology of root and shoot development revealed that the guava shoots proliferated in vitro had originated from performed meristems of the explants. Presence of BA in the proliferation medium enhanced the process of precocious axillary branching. No adventitious shoot formation was observed in any location of the explants. Roots developed from the organized tissue of the cutting base specifically just outside the vascular cylinder of the stem. Initiation of the root primordial started within 2-3 days of culture in auxin medium whereas it took 7-10 days in auxin free medium. Addition of activated charcoal in the rooting medium favoured root emergence and growth. Microcuttings rooted at 300C had their root initiation 5-15 days earlier than those rooted at 25 and 200C.
