Plantlet Regeneration from Callus Cultures in Citrus

Abastract
Callus induction and shoot regeneration was established in three rootstocks of Citrus viz. C. depressa, C. reshni and C. jambhiri. The seedlings explants (root, cotyledon, stem and leaf segments) exhibited maximum per cent callus induction upon culturing on MS medium supplemented with NAA (10 mg/1) and BAP (1 mg/1), irrespective of the genotype. Among the four explants, the stem segments exhibited maximum per cent callus induction in Citrus depressa (87.50%) and C. jambhiri (79.16%), whereas cotyledons exhibited the maximum per cent callus induction in C. reshini (80.00%). The shoot regeneration was achieved by subculturing the calli on ? MS + BAP (5 mg/1) and it was maximum is leaf-derived calli of C. depressa (42.37%), stem-derived calli of C. jambhiri (26.53%) and cotyledone derived calli of C. reshni (5.17%). In vitro raised shoots were rooted by culturing them on ? MS + IBA (2.0 mg/1) medium and the maximum per cent rooting occurred in C. jambhiri among the three genotypes. The plantlets, thus obtained were established in the pots containing loamy sand.
