In Vitro Regeneration of Multiple Shoots in Three Bamboo Species

Abastract

Plantlets of Bambusa arundinacea, Dendrocalamus brandisii and Thyrsostachys siamensis have been developed by culturing excised plumules in in vitro condition. Production of 20-75 shoots was possible by repeated transfer of cultures at 3-4 weeks? interval either in liquid or on gelled MS or B5 medium supplemented with 1.0 mg/1 BAP, the gelled medium being found better than the liquid medium. For B. arundinacea and T. siamensis, 1.0 mg/1 BAP was optimum whereas higher concentration of BAP was required for D. brandisii. Healthy roots were also produced in the same medium within 6-7 weeks after the proliferation of shoots. A large number of plantlets have been successfully transferred from aseptic condition to the outdoor nursery; they are growing satisfactorily in the field.
