Influence of Acetosyringone on Agrobacterium-mediated Transformation of Cucumber (Cucumis sativus L.)

Abastract
Genetic transformation was carried out with A. tumefaciens strain GU2260 having a binary plasmid with GUS gene containing an intron (p35SGUSINT) as a reporter and the gene for the selectable marker, neomycin phosphotransferase II (npt II). Several factors were tested to facilitate the stable integration of genes into cucumber (Cucumis sativus L.) cv. Spring Swallow genome. Acetosyringone, a phenolic compound, was shown to be essential for the transformation of cucumber and enable the stable integration of reporter gene into the cucumber genomes as shown by PCR and Southern analysis. Functional expression of the genes in transgenic plants were determined by ?-glucuronidasei (GUS) enzyme essays.
