Rapid Multiplication of Two Chrysanthemum Cultivars Through In vitro Shoot Tip Culture

Abastract

Shoot tips of Chrysanthemum morifolium Ramat cv. ?Super Yellow? and ?Light Violet? were periodically subcultured on IAA, NAA, Kn, BAP and coconut water (CW) singly or in combinations. Maximum explant regeneration was observed in both Super Yellow and Light Violet in MS supplemented with 0.8 mg/1 Kn + 2.0 mg/1 BAP + 0.5 mg/1 IAA + 0.5 mg/1 NAA + 10% CW and 0.8 mg/1 Kn + 0.5 mg/1 IAA + 10% CW, respectively. Light Violet gave highest number of multiple shoots per culture on MS supplemented with 5.0 mg/1 BAP +0.5 mg/1 NAA. Maximum callus was induced in Super Yellow on MS supplemented with 0.8 mg/1 Kn + 2.0 mg/1 BAP + 0.5 mg/1 IAA + 0.5 mg/1 NAA + 10% CW and 5.0 mg/1 BAP + 0.5 mg/1 NAA, respectively. Multiple shoot regeneration was obtained from the periodic subcultures of callus in the same medium. In in vitro regenerated shoots roots developed on all media combinations; the maximum number being found in both Super Yellow and Light Violet in half strength of MS supplemented with 0.2 mg/1 IBA and 0.2 mg/1 IAA, respectively. The rooted shoots were acclimatized and successfully transferred to plastic pots. After hardening they were transferred to the field and the survival rate was 85%. The present protocol will be helpful for rapid multiplication of some Chrysanthemums.
