Magnitude of Somaclonal Variation in Oryza sativa L. cvs. Basmati-385, JP-5, Pakhal and Swat-II

Abastract

In vitro callus formation and plant regeneration response of four rice cultvars including Basmati-385, JP-5, Pakhal and Swat-II to different culture media were studied. Two culture media one without harmones and the other with 2 mg/1 of 2,4-D and 0.2 mg/1 of kinetin were used for callus induction. Medium supplemented with growth hormones gave good results. High callus formation was observed in Pakhal (70%) and Basmati-385 (58%). For regeneration, two modified MS media, MSK2 with 2 mg/1 of Kn and 0.2 mg/1 IAA and MSK5 with 5 mg/1 Kn and 0.5 mg/1 of IAA were tested. Mean regeneration efficiency of all the genotypes on MSK5 medium was higher than that of MSK2. Genotypes were significantly different in their response to regeneration. Highest mean regeneration efficiency was recorded in JP-5 (64%) followed by Swat-II (51%), Pakhal (44%) and Basmati-385 (30%). All plants derived from tissue culture showed desirable in all the agronomic traits studied except 1000 grain weight.
