The nodal and shoot tip explants from the mature plants of Ocimum sanctum L. were cultured in vitro for proliferating the axillary shoots. Both the explants regenerated into shoots that were maintained and multiplied through periodic subculturing on half-strength MS medium supplemented with the cytokinin BA at 0.1-0.2 mg/1. Maximum frequency (91%) of in vitro multiple shoot forming culture was obtained from the mature plant explants on modified MS medium (half strength of major salts only) augmented with 0.2 mg/1 BA. Root induction in the in vitro regenerated shoots was achieved on half strength MS medium supplemented with NAA (0.1 mg/1). The plantlets were successfully transferred to soil and the percentage of survivability under ex vitro condition was 70.
