Regeneration of Plants in vitro from Seedling Explants of Pummelo (Citrus grandis (L.) Osb.)

Abastract

Using explants from in vitro grown seedlings of C. grandis on MS medium with 1/2 strength of the major salts and different concentrations of BA and kinetin cultures of proliferating axillary shoots were established. Explants from the seedlings raised on cytokinin-enriched medium responded better than those raised on cytokinin-omitted medium. Shoot proliferation efficiency of the cotyledonary node explants was 5-7 times greater than that of nodal and shoot-tip explants. At the end of three consecutive subcultures (4 week each) 30-50 shoots could be harvested from each cotyledonary node explants when cultured on medium supplemented with 1-2 mg/1 of BA. One-3-cm microcuttings prepared from in vitro proliferated >1 cm long shoots were rooted on 1/2 MS medium with both major and minor salts, and 0.1-0.6 mg/1 of either IAA, IBA or NAA included or omitted, best rooting (85%) being achieved on the medium forfeited with 0.4 mg/1 of IBA.
