In vitro Rapid Regeneration of Plantlets from Cotyledon Explant of Adhatoda vasica Nees

Abastract
Cotyledon segments from aseptically grown seedlings of Adhatoda vasica were used to initiate cultures for inducing callus and subsequent differentiation of shoot buds. Callus production was obtained within three weeks and adventitious shoot formation was observed after four weeks of incubation on MS medium supplemented with 1.0 mg/1 BA with 0.1 mg/1 NAA. Complete plantlets were obtained upon transfer of shoot cuttings on half-strength of MS salts with 0.1- 0.5 mg/1 IBA, NAA or without any growth regulator. The best and healthy rooting was observed on auxin-free medium. In vitro regenerated plantlets were transferred to potting soil and successfully established under natural condition.
