Thidiazuron-Induced Shoot Organogenesis and Plant Regeneration in Chickpea (Cicer arietinum L.)

Abastract
Direct plant regeneration from immature cotyledons via organogenesis could be obtained in three cultivars of chickpea. Thidiazuron (TDZ) either alone or in combination with auxin/antiauxin induced high frequency shoot regeneration from immature cotyledons. Shoot bud regeneration was preceded by the development of fleshy leaves (FL) which resembled the cotyledons of somatic embryos. Transfer of the cultures to fresh medium containing benzyladenine (BA, 1mg/1) and p-chlorophenoxyisobutyric acid (p-PCIB, 0.1 mg/1) resulted in the development of normal shoots. The shoots could be rooted and transferred to soil. Histological studies have confirmed the mode of regeneration as shoot organogenesis and fleshy leaves as modified leaflets.
