In vitro Direct and Indirect Regeneration of Two Cactus Species Opuntia basilaris DC. And Mammillaria elongata Engelm & Bigel

Abastract

Two species of cactus, Opuntia basilaris and Mammillaria elongata were cultured in MS supplemented with different growth regulators for in vitro direct and indirect regeneration from areoles. Optimum callus formation occurred on MS containing 3.0 mg/1 2, 4-D + 2.0 mg/1 Kn from the stem segments containing areoles of Mammillaria elongata. But callus induction was not observed in Opuntia basilaris. Maximum percentage of shoot regeneration was obtained on MS in the presence of 8.0 mg/1 2ip + 0.8 mg/1 IBA from callus in Mammillaria elongata. Highest frequency of direct regeneration of multiple shoot was observed on MS with 7.0 mg/1 BAP + 0.7 mg/1 2, 4-D in Mammillaria elongata and 6.0 mg/1 BAP + 0.05 mg/1 2,4-D in Opuntia basilaris. The plantlet of both the species were successfully established in soil.
