Regeneration of Plantlets In vitro from the Seedling Explants of Pomegranate (Punica granatum)

Abastract

Cotyledon and hypocotyl explants were collected from the in vitro grown seedlings and cultured on half-strength of MS medium for inducing callus and proliferation of adventitious shoots. The cultured explants produced callus from their cut margins and supplemently adventitious shoots on half-strength of MS medium supplemented with 0.5-0.2 mg/1 BA and 0.1-0.5 mg/1 NAA, IBA or IAA. Maximum frequency of cultures produced shoots and the highest number of shoots per culture were observed when the half-strength of MS medium was amended with 1.0 mg/1 BA and 0.1 mg/1 NAA. For rooting, the in vitro proliferated and implanted individually on root induction medium containing half-strength of MS medium supplemented with 0.5 mg/1 NAA. Within three weeks of transfer to the rooting medium 100% of the cultured microcuttings produced 2-5 roots in this medium. The rooted shoots, regenerated plantlets, were gradually acclimatized and successfully established on the soil under natural condition with about 75% survival.
