Anther Culture Response in Rice. I. Role of Strains, Adjuvants Osmoticum, Carbon Sources and Phytohormones

Abastract
Anther culture (AC) response and plantlet regeneration ability were studied in rice varieties and hybrids, involving diverse biological and physico-chemical factors. Two strains (viz. IRGC 10798 and IRGC 77130) of the same variety SR26-B displayed differential callus formation and plantlet regeneration abilities. This indicates strong involvement of genotypes (even between strains) in governing AC response. In another experiment with 100-800 mg/1 yeast extract (YE) as an organic adjuvant, 100 mg/1 induced maximum callusing and green plant regeneration. In the same variety, 200 mg/1 casien hydrolysate (CH) encouraged callusing ability, however, maximum green plantlet regeneration was observed in control. Gradual increase in CH concentration beyond 100 mg/1 exerted negative response when correlated with regeneration percentages. Mannitol as an osmoticum @ 100 mg/1 induced maximum production of androgenic calli and regenerants. As carbon source, 6% sources was found to be better than maltose and sucrose-maltose combinations on morphogenesis of androgenic calli in hybrids of IR8 ? CR 644 and BW 311-2 ? IR 52713-B-B-8-8-1-2.
