In vitro Production of Seedlings in Cymbidium aloifolium L. SW

Abastract
Seeds of Cymbidium aloifolium L. SW were aseptically cultured on four 0.8% (w/v) agar solidified basal media namely, Phytamax (PM), Modified Vacin and Went (MVW), Murashige and Skoog (MS) and Modified Knudson (MKC). Seeds germinated only on MS medium and the seedlings underwent rapid elongation when further grown on hormone-supplemented media. In terms of increase in shoot system, liquid MS + 2% (w/v) sucrose + 1.0 mg/1 NAA + 3.0 mg/1 BAP + 1.5 mg/1 Kn was the best followed by liquid MVW + 2% (w/v) sucrose + 1.0 mg/1 NAA + 3.0 mg/1 BAP + 1.5 mg/1 Kn and the others. The seedlings developed poor root system in the elongation media. A strong and stout root system developed when the seedlings were transferred on to 0.8% (w/v) agar solidified (i) half strength of MS + 1.5% (w/v) sucrose and (ii) MS + 3% (w/v) sucrose + 0.65 mg/1 IAA media within one month of culture. In respect of both number of roots developed and increase in root system half strength of MS was found to be better. Some of the in vitro grown seedlings were successfully transferred to outside pots, where they continued normal growth.
