In vitro Regeneration of Elephant Foot Yam (Amorphophallus campanulatus Blume)

Abastract
In vitro propagation of corm explants of two elephant foot yam (Amorphophallus campanulatus Blume) cultivars ?Tala? and ?Madras? has been successful. MS medium supplemented with 1.0 mg/1 NAA and 2.0 mg/1 BA was found suitable for callus induction and its maintenance. Calli were classified into corm-like structures, white translucent frible callus, pink translucent compact callus, white opaque callus and brown callus. No callus development was observed in the absence of NAA. Plantlets of cultivar ?Madras? did not develop in the medium which favoured callus development. The highest percentage of plant regeneration was observed in the medium supplemented with 0.2 mg/1 NAA and 1.0 mg/1 BA. Aging of cultures was found important for plantlet regeneration. Rate of shoot and plant regeneration under light condition was found to be higher than under dark. Plantlet survival rate was 100% when transferred to soil. No morphological variation was observed in the plantslets.
